چکیده

Nephrotic syndrome was experimentally induced by a single i.v. injection of puromyein. Permeability of the visceral epithelial wall of glomeruli was examined with electron microscope. 30-60 seconds prior to fixation, ferritin solution was injected into the urinary space of superficially located glomeruli either by direct micropuncture of the urinary space or by retrograde filling of the urinary space via the corresponding tubule. Experimental evidence is presented for formation of channel-like pathways of ferritin penetration through the epithelial layer. Since these structures in nephrotic glomeruli are surrounded by a single cell only, we would like to propose the term percellular channel for this phenomenon.

Cadherin and catenin staining in podocytes in development and puromycin aminonucleoside nephrosis.

Podocytes have been shown to express classical cadherins in the early stage of glomerulogenesis, but it remains unclear whether podocytes in normal and pathological conditions express cadherins in the adult stage. To address this question, rat podocytes in the neonate, the adult and in puromycin aminonucleoside (PAN) nephrosis were examined by immunofluorescence microscopy using antibodies agai...

Trehalose, an mTOR Independent Autophagy Inducer, Alleviates Human Podocyte Injury after Puromycin Aminonucleoside Treatment

Glomerular diseases are commonly characterized by podocyte injury including apoptosis, actin cytoskeleton rearrangement and detachment. However, the strategies for preventing podocyte damage remain insufficient. Recently autophagy has been regarded as a vital cytoprotective mechanism for keeping podocyte homeostasis. Thus, it is reasonable to utilize this mechanism to attenuate podocyte injury....

Calcineurin inhibitors cyclosporin A and tacrolimus protect against podocyte injury induced by puromycin aminonucleoside in rodent models

Podocyte injury and the appearance of proteinuria are features of minimal-change disease (MCD). Cyclosporin A (CsA) and tacrolimus (FK506) has been reported to reduce proteinuria in patients with nephrotic syndrome, but mechanisms remain unknown. We, therefore, investigated the protective mechanisms of CsA and FK506 on proteinuria in a rat model of MCD induced by puromycin aminonucleoside (PAN)...

Puromycin aminonucleoside modulates p130Cas of podocytes

PURPOSE Puromycin aminonucleoside (PAN) specifically injures podocytes, leading to foot process effacement, actin cytoskeleton disorganization, and abnormal distribution of slit diaphragm proteins. p130Cas is a docking protein connecting F-actin fibers to the glomerular basement membrane (GBM) and adapter proteins in glomerular epithelial cells (GEpCs; podocytes). We investigated the changes in...

Angiotensin type 1 receptor blocker restores podocyte potential to promote glomerular endothelial cell growth.

Both podocytes and glomerular endothelial cells (GEN) are postulated to play important roles in the progression and potential regression of glomerulosclerosis. Inhibition of angiotensin is crucial in treatment of chronic kidney disease, presumably via effects on BP and extracellular matrix. This study aimed to investigate how angiotensin inhibition altered the interactions between podocytes and...
